Oxidative Stress and Pigmentation: The Science Behind Why Spots Appear Even Without Sun

Melanin as a defense mechanism that gets out of control

Melanin is not the enemy. It is a natural protective pigment that our skin produces in response to damage. Melanocytes, specialized cells in the basal layer of the epidermis, create it through the enzyme tyrosinase, which converts the amino acid tyrosine into DOPA and then into melanin. This is then transferred to surrounding keratinocytes, where it forms a protective shield around the cell nucleus.

The skin does not appear pigmented in the sense of dark spots, but generally unhealthy, tired, without radiance. AGEs are cumulative and less reversible than other forms of oxidative damage.

Melanocyte DNA damage

The fourth pathway is the most fundamental. Oxidative stress damages the DNA of the melanocytes themselves. The cells react with an evolutionarily programmed defense reflex: they increase melanin production to protect the cell nucleus from further damage. Melanin itself is an antioxidant, so this reaction makes biological sense. From the perspective of skin aesthetics, however, this is precisely the undesirable result: locally increased pigmentation as a defensive reaction to damaged cells.

The whole chain

The mechanism of pigmentation triggered by oxidative stress can be written as a single direct sequence. Oxidative stress from UV radiation, pollution, psychological stress, or sugars damages keratinocytes. Keratinocytes send a signal to melanocytes. Melanocytes activate tyrosinase. Tyrosinase converts tyrosine to DOPA and then to melanin. Melanin is transferred to keratinocytes. Spots, uneven tone, or an overall dull skin appearance develop on the surface.

Key insight: oxidative stress is not only at the beginning of this chain as one of the triggers. It permeates the entire cascade and amplifies each step.
